Why are we doing this?
Rosewoods are the world’s most trafficked wild product. Siamese rosewoods are experiencing the most severe drought in the last three decades. Understanding how different genotypes of rosewoods might respond to drought differently will facilitate their conservation and restoration.
WP1: Leaf Area (LA) and Sample Preparation

⚠️ Lyophilised and frozen samples are very brittle. Handle with care.

❄️ Keep samples on dry ice or in –80°C freezer.

⚖️ Check the precision balance and calibrate if necessary.
</aside>
- [ ] Prepare 2 x 2 mL Eppendorf tubes with 1 stainless steel bead each. This is for Pigment and DNA samples.
- [ ] Take ALL the lyophilised leaves out from the envelope. Weigh the leaves. Put this as
All_DW.
- [ ] Pick the biggest intact leaf. Put the rest back into the envelope and keep on dry ice.
- [ ] Weigh the chosen leaf. Put this as
One_DW.
- [ ] Measure the lengths of the long axis and the short axis. Put this as
One_short, One_Long.
- [ ] Next, for each tree, we are preparing three separate samples for pigments, sugars, and DNA.
- [ ] Roughly approximate ~25 mg of leaf and weigh the sample. Put this as
Pig_DW.
